14 August 1999 Aerospray Hematology Slide Stainers Hematology Reagent Concentrate System Intended Use The SS-135/149 concentrate system is a series of concentrated hematology reagents for staining specimens with the Wescor Aerospray Hematology Slide Stainer. SS-135/149A, B, and C kits include all the ingredients you'll need to prepare standard hematology (135 series) or Basofix (149 series) stains. There will be unused items which may be discarded when making SS-135/149A or C stain (e.g., the SS-135S vial is not needed for Basofix A or C). Description Wescor reagents provide optimum performance with your Aerospray slide stainer. The reagent concentrate system delivers the quality and performance of our standard prepared reagents in a smaller package. Reagent concentrates offer lower cost, less shipping expense, and reduced storage space. You can use diluted reagent concentrates immediately after mixing. The concentrated reagent can be diluted to make SS-135 or SS-149 reagents as shown in the table on page 3. The Basofix reagents (SS-149 series) are used with the basofix quick dip fixative (SS-049P or SS-149P) to stabilize the basophil granules which otherwise dissolve in the aqueous reagents. Concentrates Diluted concentrates (SS-135/SS-149 series ) are interchangeable with prepared reagents (SS-035 and SS-049 series). As with the prepared Basofix reagents, the standard Hematology blue stain, SS-135/149 is used in both staining systems. You must also supply anhydrous methanol (SS- MeOH or equivalent) on the reagent D line for the automatic clean cycle. For Convenient mixing and storage, we recommend a 4.5 liter space-saver container and dispensing spigot for each reagent. We also offer a 4.5 liter reagent bottle assembly that allows you to connect the 4.5 liter reagent container directly to the slide stainer. These reagents contain moderately toxic chemicals that require care in handling. Always use appropriate safety measures including gloves and eye protection when handling reagents. Refer to Material Safety Data Sheets ( MSDS ) for more information. 1 Prepare Container For Basofix Pre-dip Fixative, use the 500 mL round bottle provided. For Reagents A,B, and C use a clean, empty container with 5 liter capacity. We recommend the space-saver container mentioned on page 1. If you notice any debris or precipitated reagent from previous use, clean the container before proceeding. Remove the dispensing spigot or cap and turn the container so it's spout is upright. Note: To prevent microbial growth, disinfect bulk reagent containers monthly. Fill the container with a one-tenth dilution of household bleach. Let stand for 10 minutes, then rinse throughly with deionized water. 2 Add Solvents and Concentrates Add solvents and concentrates to their containers in the order and quantities listed below: Caution! To prevent damage to the stainer, never use denatured alcohol containing ketones. Note: Do not allow the surfactant ( SS-135S ) to contact undiluted reagent concentrate- Precipitates will form. Initial and date reagent labels. Apply the appropriate label(s). Note: Before adding reagent to the 500 mL instrument bottles, check each bottle for debris or precipitated reagent. If necessary, clean the instrument bottle before refilling. To prevent microbial growth, disinfect bulk reagent containers monthly. Fill container with a one-tenth dilution of household bleach.
